How would a Western blot show differences in histone acetylation? Doesn't histone acetylation affect transcription rates? Wouldn't a Northern Blot showing differences in amount of mRNA be much more helpful?

Hi bklynite,

That's a very interesting question! You're correct that histone acetylation affects transcription, but that's a downstream effect. Conceivably we could predict that changes in acetylation would affect transcription and then try to measure transcription rates as a proxy for those changes, but that would be pretty indirect and prone to confounding, because lots of things affect transcription. Instead, it would be better to just measure acetylation in histones directly. Histones are proteins, so Western blotting would be more appropriate here. I think the lesson here may be to take the most direct approach whenever possible.
